Lecture 6: Results 1. Signal Peptide database - the Swiss-Prot Annoteted Keywords are Secreted; Signal; Stuctural protein - belongs to the actin family - length of signal sequence: 29 AA 2. SignalP - SignalP predict Sec signal peptide (Sec/SPI), Lipoprotein signal peptide (Sec/SPII), Tat signal peptide (Tat/SPI) or no signal peptide (Other) - Sec/SPI signal peptide is predicted with a probalility of 98.56%. - cleavage site between position 18 and 19: AYS-RG with a probalility of 74.8% - serum albumin (appears in the blood plasma -> secreted protein) 3. CD48 - CD48 possess a GPI-anchor and a potential GPI-modification site at position 220 was found by the analysis - outside: 225 AAs, transmembrane helix: 17 AAs, inside: 1 - yes! CD48 is an antigen and is found on the surface of several immune cells. However, CD48 does not have a transmembrane domain, but consists of a GPI anchor via a C-terminal domain. The GPI linkage is through residue 220.
